胰島素是由胰島β細胞受內源性或外源性物質如葡萄糖、乳糖、核糖、精氨酸、胰高血糖素等的激素而分泌的一種蛋白質激素。胰島素它通過促進血液中的葡萄糖被肝臟、脂肪和骨骼肌細胞吸收來調節碳水化合物、脂肪和蛋白質的代謝。 在這些組織中,吸收的葡萄糖要么通過糖化作用轉化為糖原,要么通過脂肪生成作用轉化為脂肪(甘油三酯),或者在肝臟的情況下,兩者都轉化為糖原。 血液中高濃度的胰島素會強烈抑制肝臟的葡萄糖生產和分泌。 循環中的胰島素還影響各種組織中蛋白質的合成。因此,它是一種同化激素,促進血液中的小分子轉化為細胞內的大分子。血液中的低胰島素水平具有相反的效果,它促進廣泛的分解代謝,特別是儲備的身體脂肪。β細胞對血糖水平很敏感,因此,當葡萄糖水平高時,它們會向血液中分泌胰島素;而當葡萄糖水平低時,則會抑制胰島素的分泌。胰島素能促進細胞內葡萄糖的吸收和代謝,從而降低血糖水平。它們相鄰的α細胞通過接受β細胞的提示,以相反的方式向血液中分泌胰高血糖素:血糖低時分泌增加,葡萄糖濃度高時分泌減少。胰高血糖素通過刺激肝臟中的糖原分解和葡萄糖生成來提高血糖水平。胰島素和胰高血糖素根據血糖濃度的變化而分泌入血,是葡萄糖平衡的主要機制。胰島素活性 降低或缺失會導致糖尿病,這是一種高血糖水平(高血糖)的狀況。因此,葡萄糖會在血液中積累。

分別在選取的4份健康小鼠血清、血漿、細胞培養上清中加入高濃度小鼠INS,在標準曲線動力學范圍內進行稀釋,評估線性。
